Изучение хроматографической эффективности модифицированных сорбентов Sepharose CL-4В для разделения, очистки и препаративного накопления доминирующих компонентов из экстрактов корней Hedysarum theinum Krasnob. и Hedysarum neglectum Ledeb.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. В режимах изократического и градиентного элюирования изучены сорбционная способность нативных матриц сефарозы, а также их химически модифицированного аналога. В качестве лиганда модификатора сорбента использовано производное 4-пиридинкарбоновой кислоты [(4-гидрокси-3-метоксифенил)метилен]гидразид (фтивазид). В работе рассмотрены хроматографические свойства сорбентов: Sepharose CL-4В, Sepharose CL-6В, а также азоэпоксиадсорбента Sepharose CL-4-БЭП-п-нитробензгидразид-фтивазид.</p></sec><sec><title>Результаты</title><p>Результаты. Проведенное хроматографическое исследование показало высокую эффективность применения сорбента Sepharose CL-4В и его химически модифицированного аналога в качестве носителей для жидкостной колоночной хроматографии водно-этанольных экстрактов, полученных из растений рода Hedysarum. Установлено, что полное элюирование биологически активных веществ исследуемых экстрактов достигается только с применением в качестве элюента насыщенного раствора натрия тетрабората. В полученных хроматографических фракциях определены флавоноиды и таниды различной степени конденсированности.</p></sec><sec><title>Заключение</title><p>Заключение. Сравнительное хроматографирование биологически активных веществ экстрактов корней растений рода Hedysarum на сорбентах Sepharose CL-4В и Sepharose CL-6В в равных условиях элюции показало наиболее эффективное разделение компонентов на Sepharose CL-4В. При применении химически модифицированного аналога – азоэпоксиадсорбента Sepharose CL-4-БЭПп-нитробензгидразид-фтивазид показана лучшая эффективность разделения танидов и флавоноидов. </p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Aim</title><p>Aim. To study the chromatographic efficiency of modified Sepharose CL-4B sorbents for separation, purification, and preparation of major components from Hedysarum theinum Krasnob. and Hedysarum neglectum Ledeb. root extracts.</p></sec><sec><title>Materials and Methods</title><p>Materials and Methods. Sorption capacity of the native and modified sepharose was evaluated in both isocratic and gradient elution modes. 4-pyridinecarboxylic acid derivative [(4-hydroxy-3-methoxyphenyl)methylene]hydrazide (ftivazide) was used as a sorbent modifier ligand. We studied chromatographic properties of Sepharose CL-4B, Sepharose CL-6B, and azoepoxyadsorbent Sepharose CL-4B-DEP-p-nitrobenzohydrazide-ftivazide, a chemically modified analogue of Sepharose CL-4B.</p></sec><sec><title>Results</title><p>Results. Sepharose CL-4B and Sepharose CL-4B-DEP-p-nitrobenzohydrazide-ftivazide were found efficient carriers for liquid column chromatography of water-ethanol Hedysarum extracts. Complete elution of bioactive substances has been achieved exclusively by using sodium tetraborate as an eluent. We were able to identify flavonoids and tannins of different condensation in chromatographic fractions.</p></sec><sec><title>Conclusion</title><p>Conclusion. Comparative chromatography of bioactive substances from Hedysarum root extracts is more efficient if employing Sepharose CL-4B instead of Sepharose CL-6B as a sorbent.
