Выявить наиболее целесообразные для углубленного изучения патогенетические механизмы реализации противоопухолевого и антиметастатического действия тестируемых гибридных оловоорганических соединений с помощью иммуногистохимического метода.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. В качестве тестируемого соединения было исследована субстанция бис(3,5-ди-трет-бутил-4- гидроксифенилтиолат) диметилолова (лабораторный шифр Ме-3), относящаяся к классу гибридных оловоорганических соединений. Экспериментальная часть выполнена на 30 мышах линии C57Bl/6 (самки) с использованием универсальной модели перевиваемых опухолей со спонтанным метастазированием – меланомы В16. Через 48 часов после перевивки опухолевых клеток мышам-самкам линии С57Вl/6 Ме-3 вводили внутрибрюшинно 1 раз в сутки в течение 10 дней в суммарной дозе (СД) 375 мг/кг. Для морфологического анализа был использован первичный опухолевый узел меланомы В16, подготовка тканей была проведена по стандартной схеме. Иммунофенотипирование образцов ткани меланомы В16 мышей, полученных в эксперименте, проводили с использованием первичных антител: Polyclonal Antibody to Transforming Growth Factor Beta 1 (TGFb1) (Elabscience, dilution of 1:100), Vascular Endothelial Growth Factor A (VEGFA) (Elabscience, dilution of 1:200), Polyclonal Antibody to Bcl2 Associated X Protein (Bcl2) (Elabscience, dilution of 1:100), Polyclonal Antibody to Cluster Of Differentiation 34 (CD34) Elabscience, dilution of 1:200).</p></sec><sec><title>Результаты</title><p>Результаты. В нашем исследовании под действием гибридного оловоорганического соединения (Ме-3) в опухолевой ткани активность иммуногистохимической реакции с антителами к TGF-β1 и Bcl-2 снижалась, модификация процесса образования патологических новых кровеносных сосудов и сосудоподобных структур из опухолевых клеток отмечалась только в метрономном режиме введения Ме-3 (десятикратно, в низкой дозировке).</p></sec><sec><title>Заключение</title><p>Заключение. По результатам поискового иммуногистохимического исследования ткани меланомы В16 мышей при введении гибридных оловоорганических соединений наиболее целесообразными для углубленного изучения были выбраны два патогенетических механизма реализации противоопухолевого и антиметастатического действия тестируемых соединений: апоптотический и антинеоангиогенетический. Для наиболее эффективного доклинического отбора новых перспективных соединений с предполагаемым противоопухолевым действием необходимо расширять границы применения специфических морфологических методов, в том числе и иммуногистохимических.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Aim</title><p>Aim. To identify the most suitable pathogenetic mechanisms for in-depth study of the antitumor and antimetastatic effects of tested hybrid organotin compounds using the immunohistochemical approach.</p></sec><sec><title>Materials and Methods</title><p>Materials and Methods. Here, we tested bis(3,5-di-tert-butyl-4-hydroxyphenylthiolate) dimethyltin (laboratory code Me-3), belonging to the class of hybrid organotin compounds, on 30 female C57Bl/6 mice using a universal model of transplantable tumors with spontaneous metastasis (B16 melanoma). 48 hours after tumor cell transplantation, we intraperitoneally administered Me-3 once daily to female C57Bl/6 mice for 10 days at a total dose (TD) of 375 mg/kg. For histological analysis, we used the primary tumor node of B16 melanoma. Immunophenotyping of B16 melanoma tissue samples was carried out using the polyclonal antibodies to transforming growth factor beta 1 (TGFβ-1), vascular endothelial growth factor A (VEGFA), Bcl2-associated X Protein (Bcl-2), cluster of differentiation 34 (CD34).</p></sec><sec><title>Results</title><p>Results. After the exposure to Me-3, we found a reduced immunohistochemical signal to TGF-β1 and Bcl-2 3 in the tumor tissue. Low doses of Me-3 have also impacted angiogenesis.</p></sec><sec><title>Conclusion</title><p>Conclusion.
